Biological activities

ABC294640 is a selective inhibitor of sphingosine kinase-2(SK2). The IC50s of ABC294640 against SK-HEP-1, Hep G2 and Hep 3b2.1-7 are 70, 30 and 35 μM, respectively. In human retinal endothelial cells, ABC294640 inhibits TNF-α-induced NFΚB activation with an IC50 of 20 μM which correlates with the inhibition of S1P production. In vivo, ABC294640 shows effective anti-inflammatory activity that attenuates the intestinal inflammation associated with experimental IBD in mice.[2]
