Biological activities

IPI-926 is a potent and selective inhibitor of the Hedgehog pathway.^[1] IPI-926 inhibits Gli-luciferase reporter activity at an IC50 of 9 nM in C3H10T1/2 cells transfected with WT SMO. IPI-926 shows activity against the D473H SMO mutant at an IC50 of 244 nM.^{[2]^{IPI-926 has potent, dose-dependent inhibition of Hedgehog pathway signaling and Hedgehog dependent tumor growth in a novel mouse model of medulloblastoma.^[1] In mice, IPI-926 treatment induces substantial tumor regression after 3 weeks of daily administration.^[2]}}
