Biological activities

Acadesine is a cell-permeable AMP-activated protein kinase activator which is used for the treatment of acute lymphoblastic leukemia. Acadesine may have applications in treating other disorders such as diabetes. In vitro, acadesine (0-50 µM) inhibits lipopolysaccharide (LPS)-induced expression of proinflammatory cytokines (TGF-α, IL-1, and IL-6) and inducible nitric oxide synthase in primary rat astrocytes, microglia, and peritoneal macrophages. Acadesine attenuates the LPS-induced activation of nuclear factor κB via downregulation of IκB kinase α/β activity. Acadesine also inhibits the production of inflammatory mediators in serum and their expression in CNS of rats injected with a sublethal dose of LPS by intraperitoneal injection.[1] Acadesine (0.01-0.5 mM) augments the proliferation, differentiation, and mineralization of wild-type MC3T3-E1 cells.[2] Acadesine promotes NGF-induced neurite growth and reduces neurite outgrowth inhibition in PC-12 cells treated with astroglial conditioned medium.[3]
