Biological activities

Alitretinoin is an antineoplastic agent. Alitretinoin is indicated for the treatment of skin lesions in AIDS-related Kaposi's sarcoma. Alitretinoin is a unique panagonist retinoid, capable of binding to all six known retinoid receptors (RAR-α, -β, -γ, and RXR-α, -β, -γ). [1] Alitretinoin is as potent as all-trans-retinoic acid and is more potent than 11-cis-retinoic acid and 13-cis-retinoic acid (isotretinoin) as an RAR-α activator in an in vitro study. Alitretinoin shows high affinity for the RXR receptor. [2] In vitro, alitretinoin shows more affinity to RAR than to RXR and is able to reduce the expression of inflammatory cytokines, such as tumor necrosis factor (TNF)-α, interleukin (IL)-1 β, and IL-12 p40. Alitretinoin can inhibit IL-12 p40 expression via interference with the RAR. [3]
