Biological activities

Acetaminophen is a selective cyclooxygenase-2 (COX-2) inhibitor used as a nonsteroidal anti-inflammatory drugs for the relief of headaches and other minor aches and pains. The IC50 of acetaminophen is 25.8 µM against COX-2. ^[1] Moreover, Acetaminophen is also a potent scavenger of peroxynitrite with IC50 of 72 and 11 µM against peroxynitrite-mediated phenol nitration and Sin-1-mediated DHR123 oxidation, respectively. Acetaminophen inhibits prostaglandin endoperoxide H2 synthase by eliminating the peroxide tone.^[2] In non-pretreated hamsters, single administration of acetaminophen (200-400 mg/kg i.p.) gives rise to not only elevations in serum alanine aminotransferase but also sorbitol dehydrogenase activities in a dose-related manner.^[3] Treatment with acetaminophen 10 minutes prior to formalin injection exerts a significant analgesic effect in chronic phase of formalin test.^[4]
