Biological activities

Acipimox is a niacin derivative used as a hypolipidemic agent. Acipimox inhibits the production of triglycerides by the liver and the secretion of VLDL, which leads indirectly to a modest reduction in LDL and increase in HDL. For cells isolated from normal animals, acipimox stimulates leptin release to a similar extent compared to nicotinic acid and insulin. Co-incubation of acipimox with norepinephrine or dibutyryl cAMP diminishes the stimulatory effects on leptin release, in parallel with increased lipolysis.[1] Acipimox inhibits the lipolytic rate stimulated by adenosine deaminase in a concentration-dependent manner in epididymal adipocytes isolated from Wistar rats. Acipimox also diminishes the intracellular cyclic AMP level produced by isoproterenol in the presence of adenosine deaminase in a concentration-dependent manner.[2] Acipimox may reduce triglyceride synthesis by decreasing non-esterified fatty acid availability from adipocytes. In vivo, acipimox treatment decreases basal glycerol release in subcutaneous adipose tissue.[3]
