Biological activities

Ketoprofen is a potent nonsteroidal anti-inflammatory drug. Ketoprofen has analgesic and antipyretic effects. Ketoprofen belongs to the 2-arylpropionic acid class. Ketoprofen exhibits little stereoselectivity. The absorption of ketoprofen is rapid and almost complete when given orally. Ketoprofen binds extensively to plasma albumin, apparently in a stereoselective manner.^[1] Ketoprofen (2-2000 nM) dose-dependently suppresses type II collagen synthesis associated with decreased levels of type II collagen mRNA in the absence of IL-1.^[2] The IC50 of ketoprofen is 0.13 and 0.24 µM against inhibiting the production of thromboxane B2 and PGE2, respectively.^[3] Ketoprofen depresses the synaptic responses.^[4] In addition, intravenous administration of 5 mg/kg of S(+)-ketoprofen almost completely prevents edema formation. S(+)-ketoprofen reveals a significantly antipyretic action with an ED50 of 1.6 mg/kg.^[5]
