Biological activities

Alendronate sodium, a nitrogen-containing bisphosphonate, is a potent inhibitor of bone resorption used for the treatment and prevention of osteoporosis. Alendronate sodium inhibits the isoprenoid biosynthesis pathway and interferes with protein prenylation. Alendronate sodium inhibits recombinant human farnesyl diphosphate synthase with an IC50 of 460 nM. In osteoclasts, alendronate sodium inhibits the incorporation of [3H] mevalonolactone into proteins of 18-25 kDa and into nonsaponifiable lipids, including sterols.[1] Alendronate sodium induces apoptosis in the mouse macrophage cell line J774. Alendronate sodium also inhibits murine osteoclast formation in culture.[2] In adult hound dogs experiment in vivo, locally applied alendronate sodium results in significantly increased amounts of bone in the peripheral area with both HA and TMP implants.[3] Furthermore, alendronate sodium also appears to inhibit or delay bone fusion in a rabbit model.[4]
