Biological activities

Amifostine is a cytoprotective adjuvant. Amifostine is used in cancer chemotherapy and radiotherapy involving DNA-binding chemotherapeutic agents. Amifostine is hydrolysed by alkaline phosphatase to the active cytoprotective thiol metabolite, WR-1065. The ED50 of amifostine is 255 and 230 µM against OH^- radicals at pH 7.4 and 5, respectively. Amifostine reveals scavenging activity against spontaneous lipoperoxidation.^[1] Amifostine dose-dependently prevents both platelet aggregation and TxB2 production induced by ADP, collagen and PAF.^[2] In vivo, amifostine intravenously generates an adequate radioprotective concentration.^[3]
