Biological activities

Amiloride hydrochloride is a commonly used inhibitor of Na+/H+ exchange. Amiloride hydrochloride is a potassium-sparing diuretic, used in the management of hypertension and congestive heart failure. The IC50 value for amiloride is 50 nM in the baboon bronchus and 500 nM in the rat colon. Amiloride (25 µM) inhibits the short-circuit current 21% in the rat trachea, 63% in the baboon bronchus, and 90% in the rat colon.[1] Amiloride hydrochloride inhibits TRPP3-mediated Ca2+-activated currents with IC50 value of 143 µM.[2] In psychophysical experiments, amiloride hydrochloride blocks the direct infusion of acidic solutions (pH 閳?6.0) into human skin which causes localized pain. Under more severe acidification (pH 5.0) amiloride hydrochloride is less effective in reducing acid-evoked pain.[3] Furthermore, amiloride hydrochloride potently inhibits murine tissue swelling and inflammation in response to contact sensitizing agents and ultraviolet radiation.[4]
