Biological activities

AS 1949490 is a selective SHIP2 (SH2 domain-containing inositol 5'-phosphatase 2) inhibitor. The IC50s of AS 1949490 against human and mouse SHIP2 are 620 and 340 nM, respectively. AS 1949490 also weakly inhibits human SHIP1 with an IC50 of 13 μM. In L6 myotubes, AS 1949490 increases the phosphorylation of AKT, glucose consumption and glucose uptake. In FAO hepatocytes, AS 1949490 inhibits gluconeogenesis. In vivo, acute administration of AS 1949490 suppresses the expression of gluconeogenic genes in the livers of normal mice. In diabetic db/db mice in vivo, chronic treatment with AS 1949490 significantly lowers the plasma glucose level and improved glucose intolerance.^[1]
