Biological activities

Azacitidine is a chemical analogue of the cytosine nucleoside used in DNA and RNA. Azacitidine is a potential chemotherapeutic agents for cancer. Azacitidine is used in the treatment of myelodysplastic syndrome. The IC50 values of azacitidine in six ovarian cancer cell lines (A2780, 2008/C13, Hey, NMP-1, OVCAR3, and SKOV3) range from 10.4-42.2 µM. In vitro, azacitidine pretreatment followed by 48-hour cotreatment with azacitidine and carboplatin significantly inhibits growth in 2008/C13 and Hey cells. Sequential treatment with azacitidine and carboplatin also increases cleaved caspase-3 and -8 in 2008/C13 cells.[1] Azacitidine (1-20 µM) induces apoptosis in human myeloma cell lines and primary multiple myeloma cells. Similarly, azacitidine prolongs survival from 24.5 days to 32 days in the 5T33MM murine model of systemic myelomatosis.[2] In mice bearing L1210/0 or L1210/ara-C tumors, azacitidine produces a less than or equal to 23% increase in life-span (ILS). Administration of azacitidine 3-hours after 3-deazauridine produces about an 80% ILS among mice bearing L1210/ara-C (resistant to cytarabine) tumors, but no more than an approximately 20% ILS among mice bearing L1210/0 (sensitive to cytarabine) tumors.[3]
