Biological activities

Bendamustine hydrochloride is a purine analogue/alkylator hybrid cytotoxic. Bendamustine hydrochloride shows activity against various human cancers including non-Hodgkin?? lymphoma, chronic lymphocytic leukemia, multiple myeloma, breast cancer, and small-cell lung cancer. Bendamustine hydrochloride specifically regulates, transcriptionally and posttranslationally, genes involved in apoptosis, DNA repair, and mitotic checkpoints. Bendamustine hydrochloride causes an appreciable increase in the protein expression of Bax in SU-DHL-1 cells. [1] Bendamustine hydrochloride produces DNA damage repaired via base-excision repair (BER) rather than alkylguanine-transferase mechanisms. Bendamustine hydrochloride inhibits the parental SU-DHL-1 and SU-DHL-9 cells with IC50 of 25 and 22 µM. [2] Treatment with bendamustine hydrochloride also induces a concentration-dependent apoptosis as evidenced by changes in Bcl-2 and Bax expression profiles in chronic B-cell lymphocytic leukaemia. [3] The combination of bendamustine hydrochloride with doxorubicin is strongly antagonistic on induction of apoptosis as well as on inhibition of proliferation. [4]
