Biological activities

Busulfan is an alkylating agent having a selective immunosuppressive effect on bone marrow. Busulfan has been used in the palliative treatment of chronic myeloid leukemia. Additional effects resulting from the cytotoxicity of busulfan in hematological and other tissues include lethality, sterility, teratogenicity, and alteration of immune function. Busulfan is mutagenic to microorganisms, mammalian cells in culture, Drosophila, and rodents. Busulfan is also considered potentially carcinogenic to humans.[1] In vitro, busulfan induces activin A (a member of the transforming growth factor superfamily) in a time- and concentration-dependent manner in the human cell line ECV304. Busulfan also significantly induces both tissue factor and cyclooxygenase 2. In a parallel in vivo study, busulfan treatment significantly increases serum activin A concentration.[2] In adult female mice, busulfan treatment (15, 30, or 60 mg/kg) dose-dependently increases the frequency of chromosome aberrations in bone marrow. [3]
