Biological activities

Captopril is an angiotensin-converting enzyme (ACE) inhibitor. The IC50 of captopril is 22 nM against angiotensin-converting enzyme. Minimum effective dose of captopril is 1.25 mg/kg. [1] Captopril in vitro (50閳?00 µg/mL) increases 3H-TdR incorporation in unstimulated and mitogen-stimulated cultures of rat lymphocytes. Unseparated spleen and lymph node cells of rats orally treated with captopril (50 mg/kg/day鑴?) reveals decreased basal and mitogen stimulated 3H-TdR incorporation.[2] Captopril acts directly and specifically on capillary endothelial cells, inhibiting their chemotaxis with a biphasic dose-response curve. [3] Captopril scavenges superoxide anions in vitro independent of an action on ACE.[4] Captopril enhances lymphocyte thymidine uptake induced by 25 µg/mL phytohaemagglutinin-P. Captopril results in decreased granuloma size by inhibition of angiotensin-converting enzyme in animals.[5] Captopril also inhibits angiogenesis and slows the growth of experimental tumors in rats.[3] Captopril intravenously (5 mg/kg) results in a 40% to 60% return to active shortening within 60 minutes of reperfusion. Captopril improves postischemic contractile derangements by a mechanism independent of ACE inhibition. [4]
