Biological activities

Dacarbazine is a non-classical alkylating agent, and is the mainstay of treatment for malignant melanoma. Dacarbazine exerts antitumor activities by methylation of nucleic acids or direct DNA damage, and results in arrest of cell growth or cell death. Single exposure of nonaggressive melanoma cells in culture to dacarbazine can result in overexpression of angiogenic factors such as interleukin-8 (IL-8) and vascular endothelial growth factor (VEGF), leading to increased resistance to further chemotherapy.^[1] Besides , dacarbazine inhibits the cell viability of B16-F1, A875 and SK-MEL-5 cells in a dose-dependent manner. The IC50 of dacarbazine is 259.5, 287 and 280 µM, respectively, against B16-F1, A875 and SK-MEL-5 cells. Dacarbazine inhibits normal WI-38 and L-02 cells with IC50 of 526.46 and 366.94 µM, respectively. ^[2] Dacarbazine also inhibits lung carcinoid cell line UMC-11, H727, H835 and PPC with IC50 of 23.1, 12.1, 90.6 and 47.2 µM, respectively.^[3] In vivo, dacarbazine effectively inhibits the growth of B16-F1 melanoma in C57BL/6 mice.^[2]

Protocols

Dacarbazine is dissolved in 1 M hydrochloric acid and diluted to various concentrations with culture medium. ^[2]
