Biological activities

Exjade is a rationally-designed oral iron chelator. The IC50 of exjade is 7, 8 and 31 µM against mantle cell lymphoma cells lines HBL-2, Granta-519 and Jeko-1, respectively. Exjade triggers apoptosis mediated through caspase-3 activation. Exjade decreases cyclin D1 protein levels resulting from increased proteasomal degradation. ^[1] Exjade effectively prevents cellular iron acquisition. Exjade promotes intracellular iron mobilisation. Exjade overcomes cisplatin-resistance. ^[2] Exjade monotherapy modestly prolongs survival of mice with invasive pulmonary aspergillosis. The MIC and MFC of exjade are 25 and 50 mg/L against A. fumigatus, respectively. ^[3] In human xenograft models, exjade significantly suppresses tumour growth which is associated with decreased tumour iron levels.^[2]

In vivo: Exjade is suspended in 0.5% hydroxypropylcellulose.^[2]
