Biological activities

Felbamate is a promising new antiepileptic medication. Felbamate is used for the treatment of partial onset seizures and the Lennox-Gastaut syndrome. Felbamate is also the first anticonvulsant drug with dual actions on the excitatory of NMDA and the inhibitory of GABA brain mechanisms. Felbamate (0.1-3 mM) inhibits N-methyh- aspartate (NMDA) responses and potentiates γ-aminobutyric acid (GABA) responses.[1] Slice perfusion with 1.2-1.6 mM felbamate significantly decreases the incidence of the irreversible disapperance of the CA1 electrical response induced by kainic acid. In vitro, felbamate prevents the brain damages induced in rats by ischemia or by intracerebral kainic acid injection. [2] Felbamate produces dose-dependent inhibition of [3H] TBOB binding with an IC50 of 250 µM. Felbamate also accelerates dissociation initiated by picrotoxin (PTX). Felbamate potentiates gamma-aminobutyric acid type A receptor function, at least in part, by acting at a site that interacts with the PTX site. [3]
