Biological activities

Fluocinonide is a potent glucocorticoid steroid used topically as an anti-inflammatory agent for the treatment of skin disorders such as eczema and seborrhoeic dermatitis. The penetration of fluocinonide from solution in DMSO across human abdominal skin in vitro is better in smaller volumes than in larger volumes.^[1] In a βarr2-GFP assay with WT Smo in U2OS cells, the EC50 for ?uocinonide is >5 µM. In the bodipy-cyclopamine binding assay and gli-luciferase assay, the EC50 for fluocinonide is 1 µM and 0.3 nM, respectively. In vitro, fluocinonide stimulates the proliferation of primary neuronal precursor cells.^[2] In addition, administration of stem cell factor at sites of fluocinonide administration to the dermis abolishes the mast cell-depleting effect of the glucocorticoid.^[3] In vivo, fluocinonide causes prompt and sustained pituitary-adrenocortical suppression in healthy dogs.^[4]
