Biological activities

Hydroxyurea is a M2 specific inhibitor that binds to the free radical and inactivates the enzyme ribonucleotide reductase.[1] Hydroxyurea inhibits murine leukemia L1210 cells with an IC50 of 82 µM. Hydroxyurea can reduce deoxyribonucleotide pools and increase the utilization of 3'-azido-3'-deoxythymid (ADT) in DNA synthesis. Hydroxyurea inhibits chronic myeloid leukemia (CML) cell lines K562 and RWLeu4 with IC50 of 80 and 70 µM, respectively. [2] Besides, hydroxyurea increases levels of fetal hemoglobin (HbF) and decreases morbidity from vaso-occlusive complications in sickle cell anemia (SCA). Hydroxyurea also has effects on macrocytosis and increases in the mean corpuscular hemoglobin (MCH). [3] The continuous depression of [3H] thymidmne uptake following hydroxyurea treatment is used as an indication of a G1-S block. In addition, the effects of hydroxyurea administer as a continuous infusion in vivo are dependent on both theconcentration and the duration of the infusion. In L210 tumor-bearing mice, hydroxyurea (48 mg/kg/hour) results in obvious progression delay of cells in S phase on FMF analysis. [4]

Protocols

Hydroxyurea is diluted in 0.9% NaCl solution containing heparin (20 units/mL) for i.v. continuous infusion and in 0.9% NaCl solution without heparin for i.v. push injections. [4]
