Biological activities

Indapamide is an orally active sulphonamide diuretic agent. Indapamide can reduce blood pressure by decreasing vascular reactivity and peripheral vascular resistance.^[1] 10^-4 M indapamide decreases bone resorption in organ cultures and in cocultures of osteoblast-like cells and bone marrow cells in the presence of 10^-8 M 1,25-dihydroxyvitamin D_{3 [1,25(OH)_{2D_{3]. Indapamide decreases osteoclast differentiation as the number of osteoclasts developing in coculture of marrow. Indeed, indapamide increases osteoblast-like cell proliferation and alkaline phosphatase (ALP) expression. Resorption pits produced by spleen cells cultured in the presence of soluble osteoclast differentiation factor (sODF) and CSF-1 are decreased by 10-4 M indapamide. Indapamide prevents the bone loss caused by a high-sodium diet in spontaneously hypertensive rats (SHRs) in vivo, by increasing bone formation and decreasing bone resorption without significant variations of PTH level. ^[2]}}}
