Biological activities

Irbesartan is a new long-acting non-peptide insurmountable angiotensin II AT1 receptor antagonist for the treatment of hypertension. Irbesartan displays > 10000-fold greater specificity for the AT1 receptor compared with the AT2 receptor. Irbesartan inhibits 125I-[Sar1, Ile8] - angiotensin II binding to rat kidney membranes with an IC50 of 1 nM in vitro.[1] Irbesartan also inhibits the AGE-induced tubular cell apotptosis and damage in vitro. Irbesartan inhibits the AGE-induced up-regulation of RAGE mRNA levels and subsequently reduces ROS generation in proximal tubular cells.[2] At 1 hour after dosing, irbesartan (100 mg/kg) significantly inhibits AT1 receptor binding in all anatomical areas of the kidney.[1] In vivo, irbesartan induces activation of PPARγ and improves adipose tissue dysfunction including insulin resistance.[3]
