Biological activities

JW 55 is an inhibitor of the PARP domain of tankyrase 1 and 2 (TNKS1/2). JW55 decreased auto-PARsylation of TNKS1/2 in vitro with IC50 values of 1.9 ?M and 830 nM, respectively. [1] JW 55 also decreases canonical Wnt signaling in SW480 and HCT-15 colon carcinoma cell lines. Wnt3a-induced HEK293 cells containing a transiently transfected ST-Luc (SuperTop-luciferase) reporter showed inhibition by JW55 with an IC50 value of 470 nM. In vitro, Shh Light II cells (Gli1-Luc reporter) that were activated with 50% Shh-CM were not inhibited by 10 or 1 ?mol/L JW55. Also, TNF? activated HEK293 cells containing a transiently transfected NF-?B-Luciferase (NF-?B-Luc) plasmid showed no pathway inhibition in the presence of 10 or 1 ?mol/L of JW55. [2]
