Biological activities

Lomustine is an alkylating nitrosourea compound used in chemotherapy. Lomustine is in the same family as streptozotocin. Lomustine is highly lipid soluble, and thus can cross the blood brain barrier. In vitro, lomustine triggers apoptosis in medulloblastoma and normal human epithelial and fibroblast cells through the mitochondrial pathway via decrease in the level of the anti-apoptosis proteins Bcl-2 and Bcl-xl.[1] In vivo, continued lomustine treatment (90-100 mg/m2 body surface area p.o.) results in cumulative thrombocytopenia and is effective in relapsed lymphoma.[2] Lomustine also is an active agent in the treatment of measurable mast cell tumors (MCT) in dogs. In 23 dogs with MCT, the median and mean neutrophil counts 7 days after lomustine are 1,452 cells/µL and 1,683 cells/microL, respectively.[3] Furthermore, lomustine treatment (50-110 mg/m2 body surface area p.o.) can cause delayed, cumulative dose-related, chronic hepatotoxicity that is irreversible and can be fatal in dogs.[4] In cats, lomustine also may be useful for treatment of cutaneous nonepitheliotropic lymphoma.[5]
