Biological activities

Mestinon is a parasympathomimetic and a reversible cholinesterase inhibitor. Mestinon inhibits acetylcholinesterase in the synaptic cleft, thus slowing down the hydrolysis of acetylcholine. The IC50 value of mestinon for acetylcholinesterase (AChE) in the mouse brain is 300 nM.[1] In vivo, mestinon treatment significantly decreases organophosphate induced mortality in rats exposed to the organophosphate paraoxon.[2] Mestinon treatment results in a delayed-onset, persistently exaggerated startle response in Wistar-Kyoto (WKY) rats. Treating the Wistar-Kyoto (WKY) and Sprague-Dawley (SD) rats for a second time, 7 weeks after the end of the first mestinon treatment, induces an exaggerated startle response that appears sooner and dissipated faster than is evident after the first mestinon treatment.[3] Acute administration of mestinon (10 mg/kg, by gavage) or repeated mestinon administration for 7 days (1.5 mg/kg/day, by gavage) decreases serum cholinesterase levels by approximately 50%, but neither treatment affects brain cholinesterase levels in male Sprague-Dawley rats in vivo.[4]
