Biological activities

Mycophenolate mofetil is a reversible inhibitor of inosine monophosphate dehydrogenase in purine biosynthesis (specifically guanine synthesis). Mycophenolate mofetil is an immunosuppressant and prodrug of mycophenolic acid. Mycophenolate mofetil inhibits the proliferation of lymphocytes and the production of antibodies induced by a variety of mitogens and antigens.[1] Mycophenolate mofetil inhibits mitogen-induced rat mesangial cells and human mesangial cells proliferation with IC50 value of 450 and 190 nM, respectively.[2] Mycophenolate mofetil inhibits human Tenon fibroblasts proliferation in a does-dependent manner. Brief exposure to mycophenolate mofetil leads to a reversible growth arrest for up to 14 days with concentrations of 1 mM or higher.[3] In Lewis rats in vivo, mycophenolate mofetil treatment (5, 10 or 20 mg/kg) inhibits lymphocyte proliferation and surface antigen expression in a does-dependent manner.[4] Furthermore, when given perorally at 200 mg/kg of body weight/day, mycophenolate mofetil potentiates to some extent the growth retardation induced by GCV in young NMRI mice.[5]
