Biological activities

Nevirapine is a non-nucleoside reverse transcriptase inhibitor (NNRTI). The IC50 of nevirapine is 280 nM for HIV-1 reverse transcriptase.[1] Nevirapine is used to treat HIV-1 infection and AIDS. Nevirapine is not effective against HIV-2. Resistance to nevirapine develops rapidly if viral replication is not completely suppressed.[2] The most common mutations observed after nevirapine treatment are Y181C and K103N, which are also observed with other NNRTIs.[3] Nevirapine is the most permeable antiretroviral agent compared to other HIV antiretroviral agents (such as didanosine, stavudine, zalcitabine and zidovudine) studied in the in vitro blood-brain barrier (BBB) model.[4] Nevirapine also induces extensive reprogramming of gene expression in hormone-refractory human prostate carcinoma cells in vitro. Furthermore, nevirapine down-regulates the growth of prostate carcinoma xenografts in athymic mice and induces a differentiated phenotype in vivo with increased K18 expression.[5]
