Biological activities

Nitazoxanide is a broad-spectrum drug used against intestinal parasites and bacteria.[1] Nitazoxanide is a new thiazolide antiparasitic agent that shows excellent in vitro activity against a wide variety of protozoa and helminths. [2] Nitazoxanide inhibits the 241 anaerobes (Bacteroides fragilis,Bacteroides thetaiotaomicron) with MIC90s ranging from 0.03 to 2 mg/L. The MIC90s of nitazoxanide against theBacteroides fragilis group (80 strains), Clostridium difficile (21 strains) and Clostridium perfringens (16 strains) are 0.25, 0.03 and 0.25 mg/L. [3] Nitazoxanide exhibits potent inhibition of both HBV and HCV replication with EC50 of 0.59 and 0.21 µM, respectively. Nitazoxanide is equally effective at inhibiting replication of lamivudine (LMV) and adefovir dipovoxil (ADV)-resistant HBV mutants and against 2閳?C-methyl cytidine (2閳ユmeC) and telaprevir (VX-950)-resistant HCV mutants. Nitazoxanide displays synergistic interactions with LMV or ADV against HBV, and with recombinant interferon alpha-2b (IFN) or 2閳ユmeC against HCV. Nitazoxanide induces reductions in three HBV proteins (HBsAg, HBeAg, HBcAg) produced by 2.2.15 cells with IC50 of 0.22, 0.26 and 1.1 µM, respectively. [4] In vitro treatment of E. multilocularis metacestodes with nitazoxanide induces high levels of alkaline phosphatase activity in the medium. [1]Nitazoxanide at 250 mg/kg/day significantly reduces the extent of mucosal infection in piglets. [5] Nitazoxanide has been reported to be effective against a broad range of parasites, including Giardia lamblia, Entamoeba histolytica. [2]
