Biological activities

Pomalidomide is a derivative of thalidomide that is anti-angiogenic which exerts its role as an immunomodulator. Pomalidomide markedly elevates serum IL-2 receptor and IL-12 levels, with associated activation of T cells, monocytes, and macrophages. Pomalidomide induces expression of costimulatory molecules CD28 by 48%, inducible costimulator by 48%, and inducible costimulator ligand by 66% in CD3+ gated effector cells within MM-PBMCs. Pomalidomide induces intracytoplasmic IFNγ and IL-2 expression in CD4T cells, CD8T cells and NKT cells by 2-fold, 1.9-fold, 0.8-fold, respectively.^[1] Pomalidomide abolishes or markedly inhibits the suppression of anti-CD3mAb stimulated CD25−CD4+ cell proliferation by T regulatory cells. There is a prevention of FOXP3 expression in CD25+CD4+CTLA-4+ cells up to 50% at the highest concentration of pomalidomide used (10 μM). In the CT26 murine model, pomalidomide at 50 mg/kg statistically decreases the number of CD4+/CD25+/CTLA-4/FOXP3+ T regulatory cells in the draining lymph nodes by 40% after a regimen of 6 days of treatment, compared to the PBS DMSO control.^[2]
