Biological activities

Praziquantel is a broad-spectrum anthelmintic active against schistosome species.[1]Praziquantel is also an antiparasitic drug. Praziquantel inhibits binding of [3H] ouabain to intact parasites in a pharmacologic specific manner with an IC50 of 0.9 µM. [2] The IC50 of praziquantel against three adult worms (Caenorhabditis elegans, Paramphistomum epiclitum and Schistosoma mansoni) is 484, 1.96 and 82.57 µg/mL, respectively. [3] Praziquantel can greatly enhance the anticancer efficacy of paclitaxel (PTX) in various cancer cell lines, including PTX-resistant cell lines (DLD-1 and H1299). Praziquantel synergistically enhances PTX-induced cell growth inhibition. The co-treatment of praziquantel and PTX also induce significant mitotic arrest and activate the apoptotic cascade. The cotreatment of praziquantel and PTX can markedly decrease expression of X-linked inhibitor of apoptosis protein (XIAP). Moreover, praziquantel combined with PTX results in a more pronounced inhibition of tumor growth compared with either drug alone in a mouse xenograft model. [4] Single doses of praziquantel at 50 or 100 mg/kg result in killing of all trematodes. [5]
