Biological activities

Profenal is a potent, orally effective non-steroidal anti-inflammatory drug (NSAID). Profenal has analgesic, anti-inflammatory, and antipyretic properties comparable to existing NSAIDs but with minimal gastrointestinal liabilities. In addition, profenal can inactivate human recombinant P450 2C9 in a NADPH-, time-, and concentration- dependent manner.^[1] Profenal potentiates the analgesic effects of acetaminophen and the analgesic effect is not blocked by naloxone. Profenal is an extremely potent inhibitor of arachidonate-induced writhing and is equipotent to morphine in the yeast-induced paw edema.^[2] Profenal is also effective at preventing blood-aqueous barrier disruption after paracentesis in dogs, and profenal is usefulness for treatment of prostaglandin- mediated ocular disease.^[3]

Protocols

Profenal is either suspended in 0.5% methocel or solubilized in water by the stoichiometric addition of 1 N NaOH. ^[2]
