Biological activities

RDEA-119 is a specific allosteric MEK1/2 inhibitor. RDEA-119 potently inhibits MEK1 and MEK2 in enzyme inhibition assays with IC50s of 19 and 47 nM, respectively. RDEA-119 shows broad activity at inhibiting cell proliferation in several tumor cell lines (A375 cell) in vitro. RDEA-119 inhibits MEK activity across several human cancer cell lines of different tissue origins and BRAF mutational status with EC50 ranging from 2.5 to 15.8 nM.^[1] RDEA-119 also inhibits anchorage independent growth in cell lines A375 and Colo205 at mean GI50s of 84 and 40 nM, respectively.^[2] In vivo, RDEA-119 exhibits complete suppression of ERK phosphorylation in mice.^[1]

Protocols

In vitro, RDEA-119 is dissolved in DMSO.^[1]
