Biological activities

SC144 is a first-in-class orally active small-molecule glycoprotein 130 (gp130) inhibitor.[1] SC144 shows activity in a panel of 14 human cancer cell lines from different tumor origins, with IC50s ranging from 0.14 to 4.0 μM. SC144 shows high potency in drug-resistant cancer cell lines (HEY, CRL5908, H1299 cells) with IC50s of 1.0, 3.5 and 1.67 μM, respectively.[2] SC144 induces gp130 phosphorylation (S782) and deglycosylation, abrogates Stat3 phosphorylation and nuclear translocation, and further inhibits the expression of downstream target genes.[1] In vivo in nude mice xenograft mode of huan breast cancer MDA-MB-435, SC144 (4 mg/Kg) inhibits tumor growth by 60%.[2]

In vitro, SC144 is prepared in DMSO and stored at -20 °C.[1]
