Biological activities

SCH 900776 is a selective and potent CHK1 inhibitor. The IC50 of SCH 900776 against CHK1 is 3 nM. In direct binding studies, the Kd value of SCH 900776 for the CHK1 kinase domain is 2 nM. The IC50s of SCH 900776 against CDK2 and CHK2 are 80 and 500 nM, respectively. In vitro, SCH 900776 induces dose-dependent suppression of CHK1 pS296 and concomitant accumulation of phospho-RPA signal in U2OS cells. In vivo, in combination with gemcitabine, SCH 900776 at 4 mg/kg is sufficient to induce the g-H2AX biomarker while 8 mg/kg SCH 900776 enhances tumor pharmacodynamic and regression responses relative to gemcitabine or SCH 900776 alone.[1]

Protocols

In vivo: Prior to use, SCH 900776 is dissolved in 20% hydroxypropyl β-cyclodextrin.[1]
