Biological activities

Sodium orthovanadate is an inhibitor of protein tyrosine phosphatases, alkaline phosphatases and a number of ATPases, most likely acting as a phosphate analogue. Orthovanadate inhibits the transport of dinitrophenyl-S-glutathione with an IC50 of 80 µM in human erythrocytes.[1] In transient forebrain ischemia, sodium orthovanadate as well as insulinlike growth factor-1 (IGF-1) rescues cells from delayed neuronal death in the hippocampal CA1 region.[2] Sodium orthovanadate at dosages of 10, 100 and 1000 µM can suppress the activity of alpha-glucosidase in the small intestine of normal rats, with an inhibition rate of 68.18%, 87.22% and 91.91%, respectively. Sodium orthovanadate reduces mRNA expression of alpha-glucosidase and GLUT2 in epithelial cells of small intestine.[3] In rats, a single injection of 50 mM sodium orthovanadate at 0 and 45 minutes post-middle cerebral artery occlusion (MCAO) produces similar neuroprotective effects. Furthermore, sodium orthovanadate treatment ameliorates ischemic neuronal cell injury via the activation of both protein kinase B (Akt) and extracellular signal-regulated kinase (ERK) in rats. Sodium orthovanadate also inhibits serum glucose and elicits the gradual recovery of regional cerebral blood flow (rCBF) after transient MCAO in rats. [4]
