Biological activities

Suplatast tosilate is a Th2 cytokine inhibitor. In vitro, suplatast tosilate induces a concentration-dependent suppression of purified allergen (Cry j 1)-dependent IgE synthesis in autologous B cell cultures mediated by an allergen-specific helper T cell line (SN-4), without significantly affecting the IgG synthesis. In addition, suplatast tosilate inhibits the production of interleukin 4 (IL-4) by Cry j 1-activated SN-4 as well as that by phytohemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMC) of normal donors in a concentration-dependent manner. Suplatast tosilate clearly depresses PHA-induced expression of IL-4 mRNA in normal PBMC.[1] In a mouse model of asthma in vivo, Treatment with suplatast tosilate significantly reduces the number of total cells and eosinophils in bronchoalveolar lavage fluid (BALF) (around -40%) and almost completely inhibits the development of antigen-induced bronchial hyperresponsiveness (BHR). Treatment with suplatast tosilate also significantly decreases the levels of IL-4, IL-5 and IL-13 in BALF in mice treated with suplatast tosilate compared to those in untreated mice.[2] In vivo using BALB/c mice with allergic disorders, treatment with suplatast tosilate suppresses immunoglobulin E contents in serum, and IL-4 and IL-5 levels in splenic lymphocytes.[3] Pretreatment with suplatast tosilate also prevents OVA-induced increase in IL-4 and IL-13 levels and decreases the number of lymphocytes and eosinophils in bronchoalveolar lavage fluid in bronchi of pathogen-free BALB/c mice in vivo.[4]
