Biological activities

Thiazovivin is a novel ROCK inhibitor with an IC50 value of about 0.6 µM. Thiazovivin significantly increases hESC survival after dissociation while maintaining characteristic hESC colony morphology and alkaline phosphatase (ALP) expression. Thiazovivin enhances hESCs -ECM adhesion-mediated β1 integrin signaling, which synergizes with growth factors to promote cell survival. Thiazovivin also protects hESCs in the absence of ECM by regulating E-cadherin-mediated cell-cell interaction. In addition, thiazovivin treatment significantly blocks endocytosis of E-cadherin, resulting in stabilizing E-cadherin and leading to reestablishment of cell-cell interaction, which is essential for hESC survival in ECM-free conditions. Furthermore, thiazovivin, at the concentration of 2 µM, inhibits ROCK activity and protects hESCs at a similar level as Y-27632 at the concentration of 10 µM.[1] Treatment with thiazovivin allows porcine embryonic fibroblasts (PEFs) to directly convert to fat-laden adipocytes.[2] Thiazovivin (1 µM) increases the reprogramming efficiency of CB cells to induced pluripotent stem cells (iPSCs) by more than 10 times.[3]
