Biological activities

Thymoquinone is the bioactive constituent of the volatile oil derived from the medial plant Nigella sativa. Thymoquinone has been shown to exert anti-inflammatory, anti-oxidant and anti-neoplastic effects. Thymoquinone inhibits COX-2-catalyzed PGE2 production with an IC50 of 0.3 µM. Treatment with 30 µM thymoquinone causes a 22% increase in the number of mouse papilloma cells in the G1 phase within 24 hours.^[1] Thymoquinone exhibits inhibitory effects on cell proliferation of many types of cancer cell lines, including breast adenocarcinoma, ovarian adenocarcinoma, colorectal cancer, human pancreatic adenocarcinoma, uterine sarcoma, neoplastic keratinocytes, human osteosarcoma, fibrosarcoma and lung carcinoma. Thymoquinone also significantly inhibits HUVEC invasion at 80 to 100 nM and blocks HEVEC tube formation at 100 nM. 1 µM thymoquinone significantly inhibits VEGF-induced angiogenesis, and 10 µM thymoquinone almost completely abolishes angiogenesis in the Matrigel plug assays. Thymoquinone at a dosage of 6 mg/kg/d effectively prevents the tumor growth in both size and weight in xenograft mouse model. ^[2]
