Biological activities

Tianeptine sodium salt is the sodium salt form of tianeptine. Tianeptine is a stimulator of 5-hydroxytryptamine (5-HT) uptake. Tianeptine has antidepressant effect. Tianeptine (1, 10 and 100 µM) inhibits lactate dehydrogenase (LDH) release from cortical neuronal cultures as efficiently as the N-methyl-D-aspartate (NMDA) antagonist, MK-801. Tianeptine does not block apoptosis in cultured cortical neurones caused by NMDA, but reduces apoptosis when interleukin-1β(IL-1β) is included with NMDA.[1] Tianeptine (10 mg/kg) significantly reduces both the expression of serotonin transporter mRNA and serotonin transporter binding sites labeled by [3H]paroxetine in rat dorsal raphe nucleus.[2] Ex vivo, after subchronic treatment (10 mg/kg/day i.p.) in rats, tianeptine induces an increase (30%) in [14C]serotonin uptake at a [14C]serotonin concentration of 500 nM while clomipramine induces a decrease (40%) in [14C]serotonin uptake.[3] Tianeptine (10 mg/kg) exhibits a significant antinociceptive activity in mice.[4] Furthermore, tianeptine delays the onset time of pentylenetetrazole (PTZ)-induced seizure via adenosine A1 receptors in mice.[5]
