Biological activities

Ursodiol is one of the secondary bile acids, which are metabolic byproducts of intestinal bacteria. Ursodiol is cytoprotective and has beneficial effect on the bile acid pool in the liver. In vitro, ursodiol at concentrations of 5, 25 and 50 µM inhibits the proliferation of human peripheral blood mononuclear cells in a dose-dependent manner. Ursodiol also decreases interleukin-2 production and inhibits the proliferation of the interleukin-2-dependent cell line CTLL-2.[1] In a mouse model of experimental colonic carcinoma, ursodiol inhibits the development of colon cancer.[2] In addition, supplemental dietary ursodiol (0.2% or 0.4%) is a chemopreventive agent in the azoxymethane (AOM) model of experimental colonic carcinogenesis.[3] In the AOM model of Male CD-1mice, ursodiol treatment (0.016%, 0.08%, or 0.4%) significantly inhibits the multiplicity of colonic adenocarcinoma. Ursodiol treatment also significantly lowers the proliferating cell nuclear antigen labeling index in the colonic malignancies. Ursodiol feeding reduces the expression of inducible nitric oxide synthase and tumor necrosis factor-α mRNA in the colonic mucosa.[4]
