Biological activities

Valsartan is a potent, highly selective, and orally active antagonist at the angiotensin II (AII) AT1-receptor. Valsartan is widely used for the treatment of hypertension. Valsartan competes with [125I]-AII at its specific binding sites in rat aortic smooth muscle cell membranes (AT1-receptor subtype) with a Ki of 2.38 nM.[1]Valsartan inhibits the accumulation of Aβ1-40 and Aβ1-42 in primary embryonic cortico-hippocampal neuron cultures derived from the Tg2576 mouse AD model with IC50 of 14.4 and 17.9 µM, respectively. Valsartan can attenuate oligomerization of AD-type β-amyloid protein (Aβ) peptides into high-molecular-weight (HMW) oligomeric peptides. [2] In conscious, renal hypertensive rats, valsartan induces dose-related decreases in blood pressure after both i.v. and oral administration. [1] Valsartan administration also attenuates the development of Aβ-mediated cognitive deterioration in Tg2576 mice. [2]
