Biological activities

YM-155 hydrochloride is a potent survivin suppressor. It potently inhibits survivin promoter activity with an IC50 of 0.54 nM. In vitro, YM-155, at the concentration of 10 nM, suppresses expression of survivin and induced apoptosis in PC-3 and PPC-1 human hormone-refractory prostate cancer (HRPC) cell lines. In a s.c. xenografted PC-3 tumor model in mice, continuous infusions of YM155 (3-10 mg/kg, 3 days) induces massive tumor regression accompanied by suppression of intratumoral survivin. YM155 also completely inhibits the growth of orthotopically xenografted PC-3 tumors with no significant body weight decrease. Moreover, in vivo, YM155 is highly distributed to tumors, approximately 20-fold higher concentrations than those in plasma.[1]

Protocols

In vivo: YM155 is dissolved and diluted in saline immediately before administration.[1]
