Biological activities

YM155 is a novel small-molecule surviving suppressant and potently inhibits surviving promoter with an IC50 value of 0.54 nM.[1] YM155 specifically inhibits the expression of survivin at both the mRNA and protein levels with little effect on expression levels of other IAP family or Bcl-2-related proteins. An advantage of YM155 compared with previously investigated suppressors of survivin expression is that it is active in the subnanomolar range. YM155 suppresses expression of survivin and induces apoptosis in PC-3 and PPC-1 human HRPC cell lines at 10 nM. [1] YM155 elicits cytotoxicity on glioblastoma cells with IC50s of 30-35 nM. [2] Besides that, YM155 also inhibits the survival of H460 cells with IC50 of 64 nM , Calu6 cells with IC50 of 43 nM. [3] Meanwhile, YM155 inhibits the growth of 119 human cancer cell lines, with the greatest activity in lines from non-Hodgkin?? lymphoma, hormone-refractory prostate cancer, ovarian cancer, sarcoma, non-small-cell lung cancer, breast cancer, leukemia and melanoma with the mean log growth inhibition of 50% (GI50) value of 15 nM, the mean GI50 values of YM155 are 11 nM for p53 mut/null cell lines and 16 nM for p53 WT cell lines. In the NCI-H358 xenograft model, YM155 displays superior anticancer activity to that of cisplatin or paclitaxel at highly tolerable dosage levels. [4]

YM155 at a concentration of 0-50 nM is dissolved in DMSO and incubated for 48 hours.[2]
