Biological activities

Ziprasidone is a 5-HT1A, 5-HT1B, 5-HT1D, 5-HT2A, 5-HT2C and 5-HT7 receptors antagonist. The Ki of ziprasidone is 3.4, 4, 2.3, 0.4, 1.3 and 6 nM against 5-HT1A, 5-HT1B, 5-HT1D, 5-HT2A, 5-HT2C and 5-HT7, respectively. In addition, the Ki of ziprasidone is 4.8, 7.2 and 32 nM, against D2 receptor, D3 receptor and D4 receptors, respectively. Ziprasidone is a widely used atypical antipsychotic drug. Mild concentrations of ziprasidone improve the in vitro proliferation of hippocampal-derived neural stem cells (NSCs) and increase the levels of pERK1/2 and Bcl-2 in NSCs.[1] The administration of ziprasidone significantly attenuates the immobilization stress-induced decrease in BDNF mRNA expression in the rat hippocampus and neocortex. Ziprasidone reveals differential effects on BDNF mRNA expression in the rat hippocampus and neocortex.[2] Ziprasidone attenuates the neuronal apoptosis which occurs in the hippocampus. Ziprasidone reverses the anxiety-like behaviors in rats that exposed to an enhanced single prolonged stress paradigm. Ziprasidone restores the proliferation and the protein expression of pERK1/2 and Bcl-2 in the hippocampus of these rats. [1] Acute ziprasidone at the dose of 2.5 mg/kg produces a significant attenuation of the impairment induced by sub-chronic phencyclidine in the reversal phase.[3]
