Biological activities

Zoledronate is a potent inhibitor of myeloma cell growth and secretion of IL-6 and matrix metalloproteinase-1 (MMP-1) by the tumoral environment. [1] Zoledronate is used for the treatment of multiple myeloma and controlling hypercalcemia in some malignancies and bone metastasis osteolysis. [2] Zoledronate induces both myeloma cell and bone marrow stromal cell (BMSC) apoptosis. Zoledronate significantly inhibits MMP-1 production by BMSCs stimulated with IL-1β more efficiently than pamidronate.[1] Zoledronate has proved activity in bone metastases from prostate cancer through inhibition of mevalonate pathway and of prenylation of intracellular proteins. Zoledronate synergizes with R115777 farnesyltransferase inhibitor (FTI) in inducing apoptosis and growth inhibition on epidermoid cancer cells. Zoledronate inhibits human prostate adenocarcinoma cells (PC3, LnCaP, DU145) with IC50 of 13, 18 and 34 µM, respectively. In vivo, zoledronate/R115777 combination induces cooperative effects on tumor growth inhibition of prostate cancer xenografts in nude mice with a significant survival increase. [3]
